Crystal Violet Staining

Crystal violet staining is a laboratory technique that uses a purple basic dye to visualize and classify cells, microorganisms, and biofilms, making it valuable in infection research. In Gram staining, crystal violet enters bacterial cells and forms a dye–iodine complex; alcohol or acetone removes this complex from bacteria with thinner peptidoglycan layers, while thicker-walled bacteria retain the stain. The method therefore supports rapid differentiation of Gram-positive and Gram-negative organisms. Crystal violet also binds cells attached to surfaces, allowing researchers to estimate biofilm formation by measuring the retained dye, which aids studies of microbial adhesion, persistence, and antimicrobial activity.

Here, we describe how to identify the stage of the murine reproductive (proestrus, estrus, metestrus, or diestrus) by simple, non-invasive collection and cytological assessment of vaginal smear samples. We further describe how vaginal cytology reflects circulating hormonal levels underlying transition through the murine reproductive cycle.

The intraluminal middle cerebral occlusion model in mice is herein presented. The extent of cerebral infarct is evaluated by a neurologic score and cresyl violet staining, an alternative staining to TTC, offering the great advantage to test in parallel many interest markers.
