Il-13 Expression

IL-13 expression refers to the production and regulation of interleukin-13, a type 2 cytokine that coordinates immune responses and influences tissue function. It is induced primarily in activated T helper 2 cells and innate lymphoid cells, where transcriptional programs drive IL-13 synthesis and secretion; the cytokine then signals through IL-13 receptor complexes to activate pathways such as JAK-STAT6. In immunology and infection research, measuring IL-13 expression helps characterize type 2 inflammation, host responses to parasitic infection, mucus production, and tissue remodeling. These analyses also support studies of asthma, allergic disease, immune regulation, and therapies that target IL-13 signaling.

Carbon-13 is a naturally occurring NMR-active isotope of carbon with a low natural abundance of 1.1%. In contrast, carbon-12 is the most abundant isotope of carbon with zero nuclear spin. 13C-isotope labeling is a useful technique for determining the cell central metabolism for various types of microorganisms. Two methods for assigning the α- and ε-dimethylamine nuclear magnetic resonance signals of a reductively 13C-methylated N-terminal lysine are described. One method utilizes the pH-induced selectivity of the reductive methylation reaction, and the other uses aminopeptidase to selectively remove the N-terminal lysine.

We describe an experimental setup for administrating hyperpolarized 13C-labeled metabolites in continuous perfusion mode to an isolated perfused mouse heart. A dedicated 13C-NMR acquisition approach enabled the quantification of metabolic enzyme activity in real-time, and a multiparametric 31P-NMR analysis enabled the determination of the tissue ATP content and pH.
