Dendritic Cell Pulsing

Dendritic cell pulsing is a laboratory technique that loads dendritic cells with antigen so they can present immune targets to T cells. It typically involves incubating immature or mature dendritic cells with peptides, proteins, lysates, or other antigen-containing material, allowing uptake or surface association before antigen processing and display on major histocompatibility complex molecules. In immunology and infection research, pulsed cells help assess antigen-specific T-cell activation, cytokine production, and cytotoxicity while supporting studies of pathogen immunity and vaccine design. The approach can also generate experimental cellular vaccines or test how antigen form, dose, and presentation conditions shape adaptive immune responses.

Here a protocol is presented to measure bacterial capture by CD4+ T cells which occurs during antigen presentation via transinfection from pre-infected dendritic cells (DC). We show how to perform the necessary steps: isolation of primary cells, infection of DC, DC/T cell conjugate formation, and measurement of bacterial T cell transfection.
