In Vitro In Vivo Studies

In vitro and in vivo studies are complementary research approaches that examine biological processes outside the body and within living organisms, respectively. In vitro experiments control variables such as cell type, medium, pathogen exposure, and treatment conditions, whereas in vivo studies capture interactions among immune cells, tissues, organs, and systemic responses in an intact host. In immunology and infection research, investigators use these models to characterize host-pathogen interactions, evaluate immune mechanisms and therapeutic candidates, and determine whether findings from simplified laboratory systems translate to physiological settings. Together, they improve experimental interpretation and help guide the development of vaccines, diagnostics, and anti-infective treatments.

Given the limited xenograft models available to study interactions between human CART and myeloid cells, we established in vitro and in vivo models to understand the impacts of human macrophages on CART cells. Findings can potentially be generalized to evaluate macrophage roles in the tumor microenvironment and test macrophage-targeted immunotherapies.

Clathrin-mediated endocytosis depends on adaptor proteins that coordinate cargo selection and clathrin coat assembly. Here we describe procedures to study adaptor-clathrin physical interaction and live cell imaging approaches using as a model the yeast endocytic adaptor protein Sla1p.

A primary culture of bovine corneal endothelial cells was used to investigate the mechanism of corneal endothelial-mesenchymal transition. Furthermore, a rat corneal endothelium cryoinjury model was used to demonstrate corneal endothelial-mesenchymal transition in vivo.

A simple, efficient and robust way to synchronize the delivery of multiple viral components to plant cells via Agrobacterium-mediated transient expression is described. This approach is amenable for studying replication, encapsidation followed by in vitro reassembly of non-viral components into genome depleted optical viral ghosts suitable for biomedical applications.

To study the interaction of bacteria with the blood vessels under shear stress, a flow chamber and an in vivo mesenteric intravital microscopy model are described that allow to dissect the bacterial and host factors contributing to vascular adhesion.
