Intracellular Staining

Intracellular staining is an immunological technique that uses labeled antibodies to detect proteins or other molecules inside cells, where surface measurements cannot reveal cellular function. After fixation preserves cell structure, permeabilization creates access through membranes, and fluorophore-conjugated antibodies bind specific intracellular targets for imaging or flow cytometry. In immunology and infection research, the method measures cytokines, transcription factors, signaling proteins, and pathogen components within defined cell populations. These profiles help characterize immune activation, identify infected cells, and evaluate responses to vaccines or antimicrobial treatments.

This video demonstrates the effect of tobacco product preparations, TPPs, on cytokine production from human peripheral blood mononuclear cells using flow cytometry. Treatment of the cells with TPP downregulates the production of pro-inflammatory cytokines, which is estimated after lipopolysaccharide stimulation, followed by immunostaining and flow cytometry analysis.

In this protocol a method to measure intracellular protein refolding after heat shock is described. This method can be used to study foldases like molecular chaperones and their co-factors or compounds able to influence their activity. Firefly luciferase activity is used as reporter to measure chaperone refolding activity.
