Mixed Lymphocyte Reaction

The Mixed Lymphocyte Reaction (MLR) is an in vitro immunological assay that measures how strongly lymphocytes from genetically different individuals respond to one another, making it useful for assessing cellular immune compatibility. During the reaction, antigen-presenting cells display major histocompatibility complex (MHC) molecules that allogeneic T cells recognize as foreign, triggering T-cell activation, proliferation, and cytokine production. Researchers can quantify this response using indicators such as cell proliferation or cytokine release. The MLR supports transplantation research by evaluating donor-recipient compatibility, studying alloreactivity, and investigating mechanisms involved in graft rejection and immune tolerance.

This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.
