Phenotype Determination

Phenotype determination is the identification of an organism’s observable characteristics, including cellular, molecular, and functional traits, and is essential for distinguishing biological states. In immunology and infection research, investigators combine features such as cell-surface markers, protein expression, morphology, and functional responses, often using flow cytometry, microscopy, or immunological assays to classify cells or pathogens. This approach helps identify immune-cell subsets, characterize pathogen-associated changes, and assess how infections alter host responses. Linking phenotype with genotype and disease outcome can improve understanding of pathogenesis, support diagnosis, and guide evaluation of vaccines or antimicrobial and immunomodulatory treatments.

Protocol for Plasmodium falciparum Infections in Mosquitoes and Infection Phenotype Determination

Once a gene is identified as potentially refractory for malaria, it must be evaluated for its role in preventing Plasmodium infections within the mosquito. This protocol illustrates how the extent of plasmodium infections of mosquitoes can be assayed. The techniques for preparing the gametocyte culture, membrane feeding mosquitoes human blood, and assaying viral titers in the mosquito midgut are demonstrated.

Here, we describe a method that combines in situ MHC-tetramer staining with immunohistochemistry to determine localization, phenotype, and quantity of antigen-specific T cells in tissues. This protocol is used to determine the spatial and phenotypic characteristics of antigen-specific CD8 T cells relative to other cell type and structures in tissues.

High Throughput Image-Based Phenotyping for Determining Morphological and Physiological Responses to Single and Combined Stresses in Potato

We designed an image-based phenotyping protocol to determine the morphological and physiological responses to single and combined heat, drought, and waterlogging treatments. This approach enabled the identification of early, late, and recovery responses at a whole plant level, particularly above-ground parts, and highlighted the necessity of using multiple imaging sensors.

Microfluidic flow chambers etched by photolithography and fabricated from PDMS are applied to probe functional outcomes associated with EC dysfunction and inflammation. In a representative experiment, the ability of differential shear stress to modulate monocytic cell adhesion to cytokine activated EC monolayers is demonstrated.
