Plant Defense Genes

Plant defense genes are genes that help plants detect, contain, and respond to infection or other biological threats, forming the molecular basis of plant immunity. They encode components such as pattern-recognition receptors, signaling proteins, transcription factors, and antimicrobial molecules; when receptors detect pathogen-associated molecular patterns or pathogen effectors, they activate pathways involving reactive oxygen production, cell-wall reinforcement, and defense-gene expression. In immunology and infection research, analyzing these genes clarifies how plants distinguish harmful microbes from beneficial interactions and coordinate local and systemic responses. Their study supports crop disease resistance, breeding, and strategies to reduce agricultural losses.

Quantification of pathogen growth is a powerful tool to characterize various Arabidopsis thaliana (hereafter: Arabidopsis) immune responses. The method described here presents an optimized syringe infiltration assay to quantify the Pseudomonas syringae pv. maculicola ES4326 growth in adult Arabidopsis leaves.

Robotics and Dynamic Image Analysis for Studies of Gene Expression in Plant Tissues

We report a method for introduction, tracking and quantitative analysis of GFP expression in plant cells. This method utilizes a custom-designed robotics system for semi-continuous image collection from large numbers of samples, over time. We also demonstrate the use of ImageJ and ImageReady for analysis of image series.
