Murine Periodontitis Model

The murine periodontitis model is an experimental system that reproduces key features of periodontal disease in mice, including gingival inflammation, immune activation, and alveolar bone loss. Disease is commonly induced by placing a ligature around a molar or by oral inoculation with periodontitis-associated bacteria, which promotes local microbial accumulation and triggers innate and adaptive inflammatory responses in periodontal tissues. Researchers use this model to examine host–microbe interactions, inflammatory pathways, tissue destruction, and mechanisms of bone resorption. It also supports evaluation of antimicrobial, immunomodulatory, and bone-protective therapies, while linking molecular findings to measurable clinical and histological outcomes.

This article presents a protocol for establishing a ligature-induced model of murine periodontitis involving multiple maxillary molars, resulting in larger areas of the involved gingival tissue and bone for subsequent analysis as well as reduced animal usage. A technique to assess oral neutrophils in a manner analogous to human subjects is also described.

Murine Model of Epicutaneously-Induced Immunomodulation

2025

The study demonstrates the role of the skin in modulating immune responses in mice through epicutaneous (EC) immunization. EC immunization with a protein antigen can suppress immune responses in contact hypersensitivity or reverse skin-induced suppression when combined with pathogen-associated molecular patterns. EC immunization shows promise as an effective, needle-free approach to immunotherapy.
