Saliva Harvest Technique

Saliva Harvest Technique is a standardized method for collecting oral fluid for laboratory analysis, providing a noninvasive source of immune mediators, antibodies, nucleic acids, and microbial biomarkers. The procedure typically uses passive drool or an absorbent collection device, followed by controlled handling, clarification, and storage to preserve analyte stability and reduce contamination or variability. A CD4 enumeration method, the α-test, is described which uses whole saliva to provide rapid and accurate CD4 counts. The α-test costs pennies and eliminates the need for technical training, costly reagents such as monoclonal antibodies, instrumentation, refrigeration, transport of samples, as well as collection and handling of blood.

This video demonstrates a bead-based multiplex immunoassay technique to simultaneously detect antibodies for multiple environmental pathogens in human saliva. Fluorescent beads were coupled to antigens from different pathogens, which were captured using antibodies in the human saliva sample. Upon labeling the antibodies using a fluorescent reporter, the beads were analyzed using flow cytometry to assess the presence of different pathogen-specific antibodies in the saliva.
