Sessile Mic Determination

Sessile MIC determination is a microbiological method for measuring the lowest antimicrobial concentration that inhibits surface-attached, biofilm-associated microorganisms. The assay exposes established sessile cells to serial antimicrobial dilutions and evaluates subsequent growth or metabolic activity, revealing susceptibility under biofilm conditions rather than in free-floating cultures. In immunology and infection research, this approach helps characterize how biofilm formation alters antimicrobial tolerance and persistence. Results support comparisons between planktonic and sessile susceptibility, evaluation of candidate treatments, and investigation of chronic or device-associated infections where biofilms can complicate host defense and clinical management.

Bacterial pyomelanin production results in increased resistance to oxidative stress and virulence. We report on techniques that can be used to determine inhibition of pyomelanin production and assay the resulting increase in sensitivity to oxidative stress in bacteria, as well as determine antibiotic minimum inhibitory concentration (MIC).

A protocol is presented for the convenient and high-throughput isolation and enrichment of glandular capitate stalked and sessile trichomes from Cannabis sativa. The protocol is based on a dry, non-buffer extraction of trichomes using only liquid nitrogen, dry ice, and nylon sieves and is suitable for RNA extraction and transcriptomic analysis.
