Specificity Analysis

Specificity analysis is the evaluation of how selectively an immune response, antibody, assay, or diagnostic marker recognizes its intended antigen rather than related targets. It examines molecular recognition through complementary interactions between epitopes and antibody binding sites or between peptide–MHC complexes and T-cell receptors, often by comparing reactivity across target and control antigens. In immunology and infection research, this analysis helps distinguish true pathogen-specific responses from cross-reactivity, assess diagnostic accuracy, and characterize vaccine- or infection-induced immunity. These findings support reliable serology, biomarker validation, and interpretation of immune protection.

Heterokaryon Technique for Analysis of Cell Type-specific Localization

A flexible and efficient method for the characterization of cell type-specific protein localization and nucleocytoplasmic shuttling is described. This heterokaryon approach uses fluorescently-labeled fusion proteins to image protein localizations after cell fusion. The protocol is amenable to steady-state localizations or more dynamic determinations based on live cell imaging.

A method for producing Arabidopsis leaf protoplasts that are compatible with fluorescence activated cell sorting (FACS), allowing for studies of specific cell populations. This method is compatible with any Arabidopsis line that expresses GFP in a subset of cells.

Presented here is a procedure for reproducible and statistically valid determinations of starch granule size distributions, and for specifying the determined granule lognormal size distributions using a two-parameter multiplicative form. It is applicable to all granule sizing analyses of gram-scale starch samples for plant and food science research.

An Assay for Cell Cycle Analysis of Antigen-Specific CD8+ T Cells

This video demonstrates an assay to analyze antigen-specific CD8+ T cells in different cell cycle stages. Fluorescently-labeled viable cells are selected using flow cytometry, and the cells in the different cell cycle stages are identified by their DNA content using antibodies against cell proliferation-associated nuclear protein and a nucleic acid stain.
