Elispot Method

The ELISpot method is an immunoassay that detects and quantifies individual cells releasing a specific cytokine, antibody, or other secreted biomolecule, making it valuable for measuring rare immune responses. In the assay, cells are cultured on a membrane coated with a capture antibody; secreted molecules are immobilized near each producing cell, then detected with a labeled detection antibody that forms a visible spot. Spot counts reflect the frequency of responding cells, while spot size or intensity can provide additional information about secretion. In immunology and infection research, ELISpot assesses antigen-specific T-cell and B-cell responses, vaccine immunogenicity, pathogen exposure, and immune-cell function.

Functional Assessment of Antibody-Secreting B Cells Using ELISpot Assay

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.

Optimized Interferon-gamma ELISpot Assay to Measure T Cell Responses in the Guinea Pig Model after Vaccination

The development of the interferon-gamma ELISpot assay for guinea pig PBMCs allows the characterization of T-cell responses in this highly relevant model for studying infectious diseases. We have applied the assay to measure T cell responses associated with intradermal delivery of DNA vaccines.
