Airway Mucus Rheology

Airway mucus rheology is the study of how respiratory mucus deforms and flows, providing insight into its role in airway protection and disease. Mucin polymers, water content, ions, and cellular material create a viscoelastic network whose resistance and elasticity change with applied shear, hydration, and molecular organization. Rheological measurements such as viscosity, elasticity, and yield behavior help characterize mucus clearance by cilia and coughing. In medicine, these properties support research on asthma, chronic obstructive pulmonary disease, cystic fibrosis, and airway infection, while informing the development and evaluation of therapies intended to restore mucus transport and improve respiratory function.

The viscoelastic properties of mucus play a critical role in mucociliary clearance. However, traditional mucus rheological techniques require complex and time-consuming approaches. This study provides a detailed protocol for the use of a benchtop rheometer that can rapidly and reliably perform viscoelastic measurements.

We describe a strategy to monitor maturation and migration of pulmonary dendritic cells in response to ovalbumin in the setting of ovalbumin induced allergic airway inflammation. This strategy can be modified to assess migration of pulmonary dendritic cells in settings of infection.
