Bladder Outlet Innervation

Bladder outlet innervation is the neural control of the bladder neck, urethra, and sphincter muscles that coordinates urine storage and voiding. During storage, sympathetic pathways from the thoracolumbar spinal cord help maintain outlet closure, while somatic pudendal nerves activate the external urethral sphincter; during urination, parasympathetic pathways from sacral segments promote detrusor contraction and outlet relaxation, with coordinated inhibition of sphincter activity. This integrated autonomic and somatic control enables continence and efficient emptying. Understanding bladder outlet innervation supports the evaluation of urinary retention, incontinence, neurogenic bladder, and dysfunction caused by spinal, peripheral nerve, or pelvic disorders.

Compound muscle action potential recording quantitatively assesses functional diaphragm innervation by phrenic motor neurons. Whole-mount diaphragm immunohistochemistry assesses morphological innervation at individual neuromuscular junctions. The goal of this protocol is to demonstrate how these two powerful methodologies can be used in various rodent models of spinal cord disease.

Urinary Bladder Distention Evoked Visceromotor Responses as a Model for Bladder Pain in Mice

Approximately 3-8 million people in the United States suffer from interstitial cystitis/bladder pain syndrome (IC/BPS), a debilitating condition characterized in part by pelvic pain. In order to study nervous system contributions to the condition, a physiological model of urinary bladder pain is used in mice and rats.

A Method to Target and Isolate Airway-innervating Sensory Neurons in Mice

Organ specific sensory neurons are difficult to identify. Fast Blue tracing is used to identify nodose neurons innervating the airways for cell sorting. Sorted nodose neurons are used to extract high quality ribonucleic acid (RNA) for sequencing. Using this protocol, gene expression of airway specific neurons is determined.
