Cell Sheet Transplantation

Cell sheet transplantation is a regenerative medicine technique that transfers cohesive layers of cultured cells to damaged tissue without disrupting their cell-cell connections or extracellular matrix. Cells are grown on temperature-responsive culture surfaces, then released by lowering the temperature rather than using enzymes, preserving the intact sheet and its biological properties. These sheets can be layered or applied directly to target sites, where they deliver viable cells and matrix components for tissue repair. The approach has potential in treating injuries and disease affecting epithelial, cardiac, periodontal, and other tissues, while reducing reliance on synthetic scaffolds in cell-based therapies.

Adipose-derived stem cells (ASCs) are easily isolated and harvested from the fat of normal rats. ASC sheets can be created using cell-sheet engineering and can be transplanted into Zucker diabetic fatty rats exhibiting full-thickness skin defects with exposed bone and then covered with a bilayer of artificial skin.
