Corneal Biomarkers

Corneal biomarkers are measurable molecular, cellular, or structural features of the cornea and tear film that indicate tissue health, injury, inflammation, or disease, making them valuable for clinical assessment and research. Corneal damage or altered physiology can change epithelial-cell behavior, nerve function, extracellular-matrix composition, and tear-fluid constituents, which researchers detect through imaging, sampling, and molecular assays. These indicators support the diagnosis and classification of conditions such as infection, dry eye, and corneal degeneration, while also helping monitor treatment response, predict disease progression, and evaluate tissue repair. Their development may strengthen noninvasive testing and enable more personalized approaches to ocular medicine.

Here, a step-by-step protocol for the preparation and cultivation of porcine split corneal buttons is presented. As this organo-typically cultivated organ culture model shows cell death rates within 15 days, comparable to human donor corneas, it represents the first model allowing long-term cultivation of non-human corneas without adding toxic dextran.

We describe a rat model of post traumatic stress disorder (PTSD) that reveals the persistent alterations in neuroendocrine function and the delayed long-term, exaggerated fear response, characteristic of PTSD patients. The animal model and methods described here are useful for correlating biomarkers in brain nuclei, which are mechanistic but cannot be measured in patients, with biomarkers in peripheral white blood cells, which can.

This protocol focuses on alkali burn-induced corneal neovascularization in mice. The method generates a reproducible and controllable corneal disease model to study pathological angiogenesis and the associated molecular mechanisms and to test new pharmacological agents to prevent corneal neovascularization.

Corneal Donor Tissue Preparation for Endothelial Keratoplasty

Endothelial corneal transplantation is a surgical technique for treatment of posterior corneal diseases. Mechanical microkeratome dissection to prepare tissue results in thinner, more symmetric grafts with less endothelial cell loss and improved outcomes. Dissections can be performed at the eye bank prior to corneal transplantation surgery.
