Fibroblast Matrix Remodeling

Fibroblast matrix remodeling is the process by which fibroblasts build, modify, and degrade the extracellular matrix, the structural network that supports tissues and regulates cell behavior. Fibroblasts produce collagen, fibronectin, and other matrix components, while matrix metalloproteinases and their inhibitors control degradation; during tissue injury, some fibroblasts become contractile myofibroblasts that reorganize and stiffen the matrix. In medicine, this process is central to wound healing, scar formation, organ fibrosis, and tumor progression. Understanding how fibroblasts respond to biochemical signals and mechanical forces can support the development of treatments that promote repair while limiting pathological tissue remodeling.

The aim of this method is to obtain fibroblast-derived 3D matrices as a natural scaffold for subsequent cellular assays. Fibroblasts are seeded in a pre-treated culture plate and stimulated with ascorbic acid for matrix generation. Matrices are decellularized and blocked to culture relevant cells (e.g., endothelial cells).

Nucleosome Remodeling

2020

Nucleosomes are the basic units of chromatin compaction. Each nucleosome consists of the DNA bound tightly around a histone core, which makes the DNA inaccessible to DNA binding proteins such as DNA polymerase and RNA polymerase. Hence, the fundamental problem is to ensure access to DNA when appropriate, despite the compact and protective chromatin structure. Nucleosome remodeling complex Eukaryotic cells have specialized enzymes called ATP-dependent nucleosome remodeling enzymes. These enzymes...

An experimental technique for the treatment of chondral defects in the rabbit's knee joint is described. The implantation of autologous chondrocytes seeded on a matrix is a well-accepted method for the remodeling and repair of articular cartilage lesions providing satisfying long-term results. Matrix-assisted autologous chondrocyte transplantation (MACT) offers a standardized and clinically established implantation method.
