Flux Baseline Analysis

Flux Baseline Analysis is a method for quantifying a system’s normal or background flux, providing a reference for identifying meaningful changes in measured flow, signal, or activity. It typically establishes a baseline from repeated observations under defined conditions, then compares subsequent measurements with that reference to detect deviations, trends, or abnormal variability. In medicine, this approach can support interpretation of physiological monitoring, laboratory measurements, and imaging data by separating expected background behavior from clinically relevant changes. Reliable baseline characterization improves measurement normalization, supports longitudinal assessment, and can help researchers evaluate treatment responses or disease-associated alterations.

Measuring Fast Calcium Fluxes in Cardiomyocytes

We present a method to isolate rapid (microsecond) calcium events from slower fluxes in living cells using laser scanning confocal microscopy. The method measures fluorescence intensity fluctuations of calcium indicators by recording line scans of several hundred pixels in a cell. Histogram analysis allows us to isolate the time scales of different calcium fluxes.

Metabolic reprogramming is a characteristic and prerequisite for M1 and M2 macrophage polarization. This manuscript describes an assay for the measurement of fundamental parameters of glycolysis and mitochondrial function in mouse bone marrow-derived macrophages. This tool can be applied to investigate how particular factors affect the macrophage’s metabolism and phenotype.

The presented protocol uses the eddy covariance method at non-typical locations, applicable to all types of short-canopy ecosystems with limited area, on a currently reforested windthrow site in Poland. Details of measuring site setup rules, flux calculations and quality control, and final result analysis, are described.
