High Salt Diet

A high salt diet is a pattern of eating that provides excessive sodium, an essential electrolyte whose intake can strongly influence fluid balance and cardiovascular health. When sodium accumulates in the body, it draws water into the bloodstream; the resulting expansion of blood volume can increase vascular pressure, particularly when the kidneys cannot excrete sodium efficiently. In medicine, studying high salt diet is important for understanding hypertension and related cardiovascular and renal risks, guiding dietary counseling, blood-pressure management, and public-health strategies. Research also examines how individual sensitivity, kidney function, and other dietary factors modify these effects.

The protocol describes the isolation of gut microbial EVs from salt-sensitive rats fed HSD using density gradient centrifugation. EVs were characterized by nanoparticle tracking, TEM, LPS/BCA assays, and 16S rRNA sequencing to analyze the size, morphology, composition, and microbiota origin.

A two-stage method to establish chronic kidney disease (CKD) in the Lewis rat by surgically removing 5/6th of renal mass is described. Combination of the surgical procedure, NOS-inhibition and a high-salt diet leads to a model resembling human CKD, allowing study of causal mechanisms and development of novel therapeutic interventions.

The pH of a salt solution is determined by its component anions and cations. Salts that contain pH-neutral anions and the hydronium ion-producing cations form a solution with a pH less than 7.
