Mcao Model

The middle cerebral artery occlusion (MCAO) model is an experimental method for reproducing ischemic stroke by restricting blood flow to brain regions supplied by the middle cerebral artery. In a typical procedure, researchers introduce and position an occluding filament or other obstruction within the artery, producing focal cerebral ischemia; removing the obstruction can model reperfusion injury. The model enables controlled evaluation of infarct development, neurological deficits, inflammation, blood-brain barrier disruption, and tissue recovery. In medicine, MCAO studies help clarify stroke mechanisms and assess potential neuroprotective drugs, surgical strategies, and rehabilitation approaches before clinical testing.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.

The intraluminal middle cerebral occlusion model in mice is herein presented. The extent of cerebral infarct is evaluated by a neurologic score and cresyl violet staining, an alternative staining to TTC, offering the great advantage to test in parallel many interest markers.

Embolic Middle Cerebral Artery Occlusion (MCAO) for Ischemic Stroke with Homologous Blood Clots in Rats

In this paper, we demonstrate a standard method for producing an embolic middle cerebral artery occlusion with homologous blood clots (fibrin-rich) in adult rat. This model closely mimics human ischemic stroke and is suitable for preclinical study of thrombolytic therapy for ischemic stroke.
