Non-genetic Approach

A non-genetic approach in medicine uses interventions that change disease-related processes without directly modifying a patient’s DNA or introducing genetic material. Depending on the clinical goal, these approaches act through drugs, antibodies, devices, lifestyle changes, or other treatments that alter signaling pathways, cellular activity, tissue conditions, or immune responses. They support disease prevention, diagnosis, symptom management, and therapy across many medical fields, and can provide practical alternatives when genetic therapies are unsuitable, unavailable, or still under development. Non-genetic strategies also help researchers distinguish effects caused by gene regulation from those produced by broader physiological or environmental changes.

Forward Genetic Approaches in Chlamydia trachomatis

We describe a methodology to perform genetic analysis in Chlamydia based on chemical mutagenesis and whole genome sequencing. In addition, a system for DNA exchange within infected cells is described that can be used for genetic mapping. This method may be broadly applicable to microbial systems lacking transformation systems and molecular genetic tools.

A Reverse Genetic Approach to Test Functional Redundancy During Embryogenesis

Gene function can be obscured in loss-of-function experiments if there is compensation by another gene. The zebrafish model provides a relatively high-throughput means to reveal such functional redundancy in living embryos.

Genetic Screens

2023

Genetic screens are critical tools for defining gene function and understanding gene interactions. Screens typically involve mutating genes and then assessing the affected organisms for phenotypes of interest. The process can be “forward”, where mutations are generated randomly to identify unknown genes responsible for the phenotypes, or it can be “reverse”, where specific genes are targeted for mutation to observe what phenotypes are produced. Here, JoVE reviews various types of genetic...

Reverse Genetic Approach to Identify Regulators of Pigmentation using Zebrafish

2022

Regulators of melanocyte functions govern visible differences in the pigmentation outcome. Deciphering the molecular function of the candidate pigmentation gene poses a challenge. Herein, we demonstrate the use of a zebrafish model system to identify candidates and classify them into regulators of melanin content and melanocyte number.

An Overview of Genetics and Disease

Many human diseases are associated with mutations or variations in genetic sequences. Some of these genetic variants are heritable, passed down from generation to generation, while others arise sporadically during an organism’s life and cause diseases such as cancer. Researchers are trying to identify and characterize these genetic alterations in the hopes of improving diagnosis and therapeutic options for patients. In this video, we will examine the history of genetic disease research, and...
