Retinal Reactive Gliosis

Retinal reactive gliosis is a cellular response in which retinal glial cells, particularly Müller glia and astrocytes, alter their structure and function after injury, disease, or environmental stress. Damage-associated signals trigger glial hypertrophy, changes in gene expression such as increased glial fibrillary acidic protein, and the release of inflammatory mediators that can support tissue repair but may also disrupt neuronal function and form a glial scar. In medicine, studying retinal reactive gliosis helps clarify how conditions such as diabetic retinopathy, glaucoma, retinal detachment, and neurodegeneration progress. Its molecular markers and pathways may guide disease monitoring and therapies designed to preserve retinal neurons and vision.

This article describes a detailed protocol to produce a forebrain stab injury in adult mice. The stab injury induces severe reactive gliosis and glial scar formation which can be subsequently examined by standard immunohistochemistry methods.

Preparation Steps for Measurement of Reactivity in Mouse Retinal Arterioles Ex Vivo

Many vision-threatening ocular diseases are associated with dysfunctional retinal microvessels. Therefore, the measurement of retinal arteriole responses is important to investigate the underlying pathophysiological mechanisms. This article describes a detailed protocol for mouse retinal arteriole isolation and preparation to assess the effects of vasoactive substances on vascular diameter.

Modeling Retinal Degeneration and Regeneration in Zebrafish Using a Focal Laser Injury

2025

This video demonstrates a procedure to induce retinal degeneration in adult zebrafish using laser injury. The laser targets photoreceptor cells in the retina, triggering cell death and a degenerative process. This damage activates Müller glia (MG) cells, leading to their dedifferentiation into stem cell progenitors that aid in retinal regeneration.

Detecting Reactive Oxygen Species

2023

Reactive oxygen species are chemically active, oxygen-derived molecules capable of oxidizing other molecules. Because of their reactive nature, there are many deleterious effects associated with unchecked ROS production, including structural damage to DNA and other biological molecules. However, ROS can also be mediators of physiological signaling. There is accumulating evidence that ROS play significant roles in everything from activation of transcription factors to the mediation of...
