Rna Removal Step

An RNA removal step is a nucleic-acid purification stage that eliminates RNA contamination from a DNA preparation, improving the accuracy and consistency of downstream analysis in medicine and biomedical research. It commonly uses RNase, an enzyme that hydrolyzes RNA into smaller fragments, followed by precipitation, washing, or column-based cleanup to separate the digested material from purified DNA. Removing residual RNA helps produce cleaner genomic DNA for PCR, sequencing, genotyping, and other molecular diagnostic workflows, where contaminating nucleic acids can affect concentration measurements, sample quality, or interpretation. The step is therefore important for reliable clinical and research results.

Regulation of Expression Occurs at Multiple Steps

2020

Gene expression can be regulated at almost every step from gene to protein. Transcription is the step that is most commonly regulated. This involves the binding of proteins to short regulatory sequences on the DNA. This association can either promote or inhibit the transcription of a gene associated with the respective sequence. Transcription results in the generation of precursor (pre-mRNA) that consists of both exons and introns, which needs further processing before being translated to a...

RNA-Seq

Among different methods to evaluate gene expression, the high-throughput sequencing of RNA, or RNA-seq. is particularly attractive, as it can be performed and analyzed without relying on prior available genomic information. During RNA-seq, RNA isolated from samples of interest is used to generate a DNA library, which is then amplified and sequenced. Ultimately, RNA-seq can determine which genes are expressed, the levels of their expression, and the presence of any previously unknown transcripts.

2016

Retinal pigment epithelium (RPE) replacement strategies and gene-based therapy are considered for several retinal degenerative conditions. For clinical translation, large eye animal models are required to study surgical techniques applicable in patients. Here we present a rabbit model for subretinal surgery geared towards RPE transplantation, which is versatile and cost-efficient.

Here, we detail the method of Sequencing of Psoralen crosslinked, Ligated, and Selected Hybrids (SPLASH), which enables genome-wide mapping of intramolecular and intermolecular RNA-RNA interactions in vivo. SPLASH can be applied to study RNA interactomes of organisms including yeast, bacteria and humans.
