Breakpoint Response Measurement

Breakpoint response measurement quantifies the maximum effort an animal will exert to obtain a reinforcing outcome, providing a behavioral index of motivation and reward value in neuroscience. In a progressive-ratio procedure, the response requirement increases after each earned reward until the subject stops responding; the breakpoint is the highest completed requirement. Researchers use this measure to assess how neural circuits, pharmacological treatments, brain lesions, or environmental conditions alter reward-driven behavior. Comparing breakpoint values helps distinguish changes in motivation from changes in motor capacity or general activity, supporting research on reinforcement, addiction, and disorders involving altered reward processing.

Source: Lee, D., et. al. Modified Experimental Conditions for Noise-Induced Hearing Loss in Mice and Assessment of Hearing Function and Outer Hair Cell Damage. J. Vis. Exp. (2023)This video demonstrates the measurement of auditory brainstem response (ABR) in a mouse. Electrodes are placed on the head, behind the ear, and near the tail of an anesthetized mouse. Acoustic stimuli at various intensities are delivered, and electric signals generated are recorded as waveforms.

Measurement of Compressive Stress-Strain Response at Small-Strains

This protocol presents the configuration of a compression test device capable of precisely measuring the mechanical properties of microstructures in the small-strain region, along with a systematic method for compression testing using this device. The proposed device can be used to analyze various microstructures and the mechanical behavior of polymer-based materials.

To understand a link between the immune response and behavior, we describe a method to measure locomotor behavior in Drosophila during bacterial infection as well as the ability of flies to mount an immune response by monitoring survival, bacterial load, and real-time activity of a key regulator of innate immunity, NFκB.

We developed the Visual-Motor Response to quantitate the motor output of larval zebrafish in response to light increments and decrements. We also examined zebrafish vision mutants, including the no optokinetic response (nrc) mutants, which were thought to be completely blind when tested by another vision assay, the optokinetic reflex.
