Carbon Fiber Microelectrodes

Carbon fiber microelectrodes are miniature electrochemical sensors made from conductive carbon fibers and used to measure chemical signaling with high spatial and temporal resolution. In neuroscience, a potential applied to the fiber drives oxidation or reduction of electroactive molecules at its surface, generating a current proportional to analyte concentration. These electrodes support fast-scan cyclic voltammetry and amperometry for monitoring neurotransmitters such as dopamine in living brain tissue, often alongside electrophysiological recordings. Their small size helps reduce tissue displacement while enabling measurements in specific neural regions, making them valuable for studying synaptic communication, reward circuits, disease mechanisms, and responses to pharmacological treatments.

This protocol describes a procedure for constructing carbon fiber microelectrode arrays for chronic and acute in vivo electrophysiological recordings in mouse (Mus musculus) and ferret (Mustela putorius furo) from multiple brain regions. Each step, following the purchase of raw carbon fibers to microelectrode array implantation, is described in detail, with emphasis on microelectrode array construction.

In this video, Drosophila larvae nerve fibers are immersed in FM dye, which binds to cell membrane lipids. Suction microelectrode and electrical pulses stimulate the endocytosis process, incorporating the FM dye into newly formed endocytic vesicles. This method, which visualizes presynaptic stages of neurotransmission, is useful for studying synaptic function and neurotransmitter release.
