Dauer Pheromone

Dauer pheromone is a chemical signal produced by nematodes that communicates unfavorable environmental conditions, making it an important model for studying how sensory cues shape neural and developmental decisions. In Caenorhabditis elegans, pheromone components called ascarosides accumulate with population density and limited resources, activating sensory neurons and neuroendocrine pathways that suppress reproductive development and promote entry into the stress-resistant dauer larval stage. Research on this signal links olfactory detection to hormone-like signaling, metabolism, behavior, and aging. It also provides a tractable system for investigating how nervous systems integrate external information to coordinate organism-wide physiological responses.

In mice, the ability to detect pheromones is principally mediated by the vomeronasal organ (VNO). Here, an acute tissue slice preparation of VNO for performing calcium imaging is described. This physiological approach allows observations of subpopulations and/or individual neurons in a living tissue and is convenient for receptor-ligand identification.
