Epileptogenic Focus Localization

Epileptogenic focus localization is the process of identifying the brain region that initiates epileptic seizures, an essential step in diagnosing drug-resistant epilepsy and planning treatment. Clinicians integrate seizure history, neurological examination, scalp or intracranial electroencephalography, and imaging such as MRI, positron emission tomography, or single-photon emission computed tomography to determine where abnormal electrical activity begins and how it spreads. By defining the seizure onset zone and distinguishing it from surrounding regions involved during a seizure, this approach supports surgical evaluation, targeted ablation, and other personalized interventions while helping protect eloquent brain areas responsible for language, movement, and memory.

This video describes a method of temporarily disrupting the blood-brain barrier (BBB) by creating transient openings using microbubble-mediated focused ultrasound (FUS). We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.
