Ev Separation

Extracellular vesicle (EV) separation is the process of isolating membrane-bound particles released by cells from complex biological fluids, enabling their study as carriers of molecular information. Separation methods exploit differences in vesicle size, density, surface markers, or physicochemical properties and include differential ultracentrifugation, size-exclusion chromatography, precipitation, and affinity-based capture. In neuroscience, purified EV populations can be analyzed for proteins, lipids, and nucleic acids involved in cell-to-cell communication, neuronal function, and disease mechanisms. Reliable separation supports biomarker discovery, investigation of neurodegenerative disorders, and development of EV-based strategies for diagnosis or therapeutic delivery, while careful method selection helps preserve vesicle integrity and reduce sample contamination.

Chimeric antigen receptor (CAR) cell-derived EVs (CAR-EVs) isolated using ion-exchange chromatography exhibit increased cargo capacity, significantly enhancing their functional efficacy. This study further characterizes CAR-EVs to elucidate their biological activity and therapeutic potential.
