Fly Sedation Assay

The fly sedation assay is a behavioral method that measures how Drosophila respond to sedative or anesthetic exposure, providing a practical model for studying nervous-system function. In a typical assay, flies receive a controlled dose of an agent, and researchers record sedation through reduced movement, loss of coordinated behavior, or failure to recover over time; differences in onset and recovery can reflect genetic, neural, or pharmacological effects. This approach supports neuroscience research on neural circuits, neurotransmitter systems, drug sensitivity, and mechanisms that regulate arousal and behavioral recovery.

Straightforward assays for measuring ethanol sensitivity and rapid tolerance in Drosophila facilitate the use of this model organism for investigating these important ethanol-related behaviors. Here, a relatively simple, scalable assay for measuring ethanol sensitivity and rapid tolerance in flies is described.

This video describes a simple method to quantify fly locomotion and shows the climbing assay performed. The fly climbing assay is a general test of motor function which relies on a negative geotaxis response.

Drosophila Aggression Assay: A Method to Test Behavioral Plasticity in Flies

2023

Aggression is an innate behavior and previous fighting experience affects fighting strategy and outcome of later contests in flies. This video describes an assay used to study Drosophila's aggressive behavior. The example protocol features the assay performed in a setup that eliminates fly handling and therefore reduces stress for the animals, which would otherwise impact the subjects' performance.
