Map Two Immunoreactivity

MAP2 immunoreactivity is an antibody-based method for detecting microtubule-associated protein 2, a neuronal cytoskeletal protein that is particularly enriched in dendrites. In this approach, tissue or cultured cells are fixed, exposed to primary antibodies that bind MAP2 epitopes, and visualized with labeled secondary antibodies, producing a signal that reveals MAP2 distribution. In neuroscience, MAP2 immunoreactivity helps identify neurons, distinguish dendritic from axonal compartments, and assess neuronal morphology, maturation, and structural changes after injury or experimental treatment. Quantifying staining patterns can support studies of dendritic architecture, neuronal development, and neurodegenerative processes.

Here we present a protocol to visualize spatial correlation of calcitonin gene-related peptide (CGRP)-immunoreactive nerve fibers and blood vessels in the cranial dura mater using immunofluorescence and fluorescent histochemistry with CGRP and phalloidin, respectively. In addition, the origin of these nerve fibers was retrograde traced with a fluorescent neural tracer.
