Microglia Activation

Microglia activation is the process by which the brain’s resident immune cells shift from homeostatic surveillance to responsive states after detecting infection, injury, protein aggregates, or other disturbances in neural tissue. Signals from damage- or pathogen-associated molecules engage microglial receptors, triggering changes in morphology, motility, gene expression, cytokine release, phagocytosis, and antigen presentation; the response can also resolve and return toward homeostasis. Studying microglia activation helps explain neuroinflammation and its roles in stroke, traumatic injury, Alzheimer’s disease, and other neurological disorders, while informing biomarker development and strategies to limit harmful inflammation without disrupting protective immune functions.

Microglia (immune cells of the brain), are used as a surrogate biosensor to determine how nanoparticles influence neurotoxicity. We describe a series of experiments designed to assay microglial response to nanoparticles and exposure of hypothalamic neurons to supernatant from activated microglia to determine neurotoxicity.

We outline methods for the efficient and quick isolation/culture of viable microglia from the neonatal cerebral cortex and adult spinal cord. The dissection and plating of cortical microglia can be accomplished within 90 minutes, with the subsequent microglial harvest taking place ~ 10 days following the initial dissection.
