Mouse Brain Microglia

Mouse brain microglia are resident immune cells of the central nervous system that support neural health and provide a tractable model for studying neuroinflammation and brain disease. They continuously survey surrounding tissue with motile processes, detect injury or altered signals, and change their gene expression, morphology, and behavior to engulf cellular debris or release inflammatory mediators. In neuroscience, researchers isolate, image, or genetically manipulate these cells to examine interactions with neurons, synapses, and other immune cells. These studies clarify how microglial responses influence development, infection, injury, neurodegeneration, and therapeutic outcomes, linking cellular mechanisms to whole-brain function.

Magnetic Isolation of Microglia from Mouse Pup Brains

2025

In this video, we describe a protocol for isolating CD11b cells, including microglia, from mouse pup brains using magnetic-activated cell sorting. The brain tissues are homogenized to obtain single-cell suspensions, which are incubated with anti-CD11b antibody-coated microbeads, followed by a magnetic column-based separation to purify the CD11b+ cell population.

Isolation of Microglia from the Cortex of an Adult Mouse Brain

2025

The video demonstrates a technique for isolating microglia from the cortex of an adult mouse brain. First, the cortex undergoes segmentation and homogenization to remove neurons and other glial cells, preserving microglia. Following that, the cell suspension is exposed to anti-myelin magnetic microbeads and a magnetic field, effectively separating microglia and myeloid cells from myelin debris and cells containing myelin.

Isolating Microglia from Mouse Brain Demyelinating Lesions Using Magnetic Activated Cell Sorting

2025

This video demonstrates a protocol for extracting microglial cells from demyelinating lesions in mouse brains using magnetic cell sorting. The protocol involves dissecting the lesioned brain tissue containing microglial cells, treating the tissue with specific enzymes to obtain a single-cell suspension, and utilizing microglial receptor-specific magnetic beads to isolate the microglial cells.

Isolation and Culture of Microglia from Rat Brains using Serum-Free Media

Source: Bosco, A., et al. In Vivo Dynamics of Retinal Microglial Activation During Neurodegeneration: Confocal Ophthalmoscopic Imaging and Cell Morphometry in Mouse Glaucoma. J. Vis. Exp. (2015) This video demonstrates in vivo imaging of retinal microglia in a mouse model of glaucoma. A transgenic mouse with fluorescent protein-expressing retinal microglia is positioned under an ophthalmoscope. Infrared illumination is used to locate the optic nerve head (ONH), and the focal plane is...
