Mouse Coronal Slicing

Mouse coronal slicing is a neuroanatomical technique that cuts the brain into cross-sectional slices perpendicular to its rostrocaudal axis, allowing researchers to examine regional structure and organization. After the brain is removed and stabilized, a vibratome or microtome advances a blade through the tissue at controlled thickness and speed, producing serial sections for staining, imaging, or electrophysiological analysis. These slices preserve relationships among regions such as the cortex, hippocampus, and thalamus, supporting studies of neural circuits, development, disease, and injury. Coronal sections also provide a standardized framework for comparing experimental findings with brain atlases and assessing cellular or molecular changes.

In mice, the ability to detect pheromones is principally mediated by the vomeronasal organ (VNO). Here, an acute tissue slice preparation of VNO for performing calcium imaging is described. This physiological approach allows observations of subpopulations and/or individual neurons in a living tissue and is convenient for receptor-ligand identification.
