Neurochemical Injection

Neurochemical injection is a research method that delivers a defined neurotransmitter, neuromodulator, or related compound into a selected site to examine how chemical signaling influences nervous-system function. The injected substance acts locally or enters nearby circuits, where it can bind receptors, alter neuronal excitability, and modify synaptic transmission under controlled dose and timing conditions. In neuroscience, this approach helps link particular chemicals to behaviors, physiological responses, and disease-related changes, while supporting studies of neural pathways and pharmacological mechanisms. Careful targeting, delivery controls, and comparison with appropriate control injections are essential for determining whether observed effects arise from the compound, its location, or the injection procedure.

This protocol presents a method for the morphological recovery of neurons patched during electrophysiological recordings using biocytin filling and subsequent immunohistochemical postprocessing. We show that thick biocytin-filled sections that were stained and coverslipped can be restained with a second primary antibody days or months later.
