Striatum Dissection

Striatum dissection is a neuroanatomical technique used to isolate the striatum, a group of subcortical nuclei involved in movement, action selection, learning, and reward. The procedure relies on recognizing the striatum’s position and boundaries within the brain and carefully separating its tissue from neighboring regions, often in freshly collected brain slices. Isolated striatal samples can then support molecular, cellular, electrophysiological, or histological analyses of neural circuits and neurotransmitter systems. This approach helps researchers investigate basal ganglia function, model neurological and psychiatric disorders, and assess how drugs, genetic changes, or neural injury alter striatal structure and activity.

This protocol describes in detail how to specifically transfect different regions in the C57BL/6 central nervous system via in utero electroporation. Included in this protocol are detailed instructions for transfections of regions that develop into the cortex, hippocampus, thalamus, hypothalamus, lateral septal nucleus and striatum.

Explants from the midbrain dopamine system and striatum are used in a collagen matrix assay for the in vitro analysis of mesostriatal and striatonigral pathway development. In this assay axonal outgrowth and guidance can be manipulated and quantified. It can also be modified for assessing other regions or molecular cues.

This video presentation shows a method of harvesting the two most important highly vascular structures that support forebrain function. They are the cerebral surface (superficial) vasculature along with associated meninges (MAV) and the choroid plexus which are necessary for cerebral blood flow and cerebrospinal fluid (CSF) homeostasis.
