Neurogenin 2 Neurons

Neurogenin 2 neurons are induced neuronal cells generated by directing progenitor or pluripotent stem cells toward a neuronal fate through expression of the transcription factor Neurogenin 2 (NGN2). NGN2 activates a neurogenic gene program that promotes cell-cycle exit, neuronal differentiation, and development of functional, typically excitatory neurons, often through a rapid and reproducible in vitro process. In neuroscience, these cells provide a scalable model for studying neuronal development, synapse formation, network activity, and disease mechanisms, while supporting drug screening and functional testing. Their defined generation can also improve experimental consistency compared with some primary neuron preparations.

Calcium signals play a key role in many cellular processes including gene expression, survival and differentiation. Here we demonstrate how to perform calcium imaging using Fura-2 AM. Calcium imaging is a valuable tool to study the regulation of intracellular calcium in real time and its regulation of signaling cascades.

Fluorescence and Bioluminescence Imaging of Subcellular Ca2+ in Aged Hippocampal Neurons

Intracellular Ca2+ remodeling in aging may contribute to excitotoxicity and neuron damage, processes mediated by Ca2+ overload. We aimed at investigating Ca2+ remodeling in the aging brain using fluorescence and bioluminescence imaging of cytosolic and mitochondrial Ca2+ in long-term cultures of rat hippocampal neurons, a model of neuronal aging.

Source: Josiah, S. S., et al. Study of the Functions and Activities of Neuronal K-Cl Co-Transporter KCC2 Using Western Blotting. J. Vis. Exp. (2022).The video demonstrates the steps for detecting and quantifying phosphorylated potassium-chloride cotransporter 2 (KCC2) protein in a cell lysate. This process involves immunoprecipitation to isolate phosphorylated KCC2 and chemiluminescence to quantify the isolated protein.
