Neuronal Firing

Neuronal firing is the process by which neurons generate and transmit electrical signals, enabling communication throughout the nervous system. When membrane depolarization reaches threshold, voltage-gated sodium channels open and drive an action potential, followed by potassium channel activation that repolarizes the membrane; ion gradients are then restored to prepare the neuron for subsequent activity. The timing and frequency of neuronal firing encode information and influence synaptic communication, sensory processing, movement, and behavior. Studying firing patterns helps neuroscientists understand neural circuits and investigate how altered excitability contributes to neurological disorders.

Inducing Plasticity of Astrocytic Receptors by Manipulation of Neuronal Firing Rates

Here we describe an adaptation of protocols used to induce homeostatic plasticity in neurons for the study of plasticity of astrocytic G protein-coupled receptors. Recently used to examine changes in astrocytic group I mGluRs in juvenile mice, the method can be applied to measure scaling of various astrocytic GPCRs, in tissue from adult mice in situ and in vivo, and to gain a better appreciation of the sensitivity of astrocytic receptors to changes in neuronal activity.

The video demonstrates an assay to identify target-selective neurons via simultaneous eye-tracking and single-neuron recordings. A human participant is tasked with identifying a target object among an array of objects in a display, and the eye movement is tracked. Simultaneously, single neurons' firing rate is recorded via electrodes implanted in the brain. The data from both recordings are correlated to assess the presence of target-selective neurons.
