Photostimulation-assisted Identification

Photostimulation-assisted identification is a neuroscience method that uses targeted light to identify neurons or neural connections during experimental recordings. In practice, light-sensitive proteins such as opsins are activated in selected cells, and resulting electrical or synaptic responses are measured to determine whether recorded neurons belong to, or receive input from, the stimulated population. By linking cellular activity to genetically defined or anatomically localized circuits, the approach helps researchers map connectivity, distinguish neuronal subtypes, and test causal relationships between circuit elements and behavior. It is valuable in studies of sensory processing, motor control, learning, disease mechanisms, and the organization of complex neural networks.

Mapping Inhibitory Neuronal Circuits by Laser Scanning Photostimulation

This paper introduces an approach of combining laser scanning photostimulation with whole cell recordings in transgenic mice expressing GFP in limited inhibitory neuron populations. The technique allows for extensive mapping and quantitative analysis of local synaptic circuits of specific inhibitory cortical neurons.
