Serum-free Culture

Serum-free culture is a cell culture method that grows cells without animal serum, enabling more controlled and reproducible experimental conditions. Instead of relying on the complex, variable mixture of proteins, lipids, hormones, and growth factors in serum, researchers supply defined nutrients and supplements selected for the cell type and study objective. In neuroscience, this approach supports the maintenance, differentiation, and analysis of neurons, neural stem cells, and glial cells while reducing background signals that can obscure cellular responses. Serum-free systems therefore improve studies of neural development, cell signaling, disease mechanisms, and responses to candidate therapeutics.

This protocol describes in detail the method for generating neural progenitors from embryonic stem cells using a serum-free monolayer method. These progenitors can be used to derive mature neural cell types or to study the process of neural specification and is amenable to multiwell format scaling for compound screening.

An Assay to Detect Autoantibodies in Human Serum using a Hippocampal Neuronal Culture

This video demonstrates a technique for identifying antibodies against specific antigens in a sample using a rat hippocampal neuronal culture. By exposing cultured neurons to patient samples containing autoantibodies targeting N-Methyl-D-aspartate receptors (NMDARs), the presence of autoantibodies bound to the neuronal surface is revealed through immunostaining.

Serum utilized in embryo cultures contains unknown components that can affect the outcome of experiments especially in studies involving signaling interactions. Here we utilized a serum-free oxygenated culture system and show that mid-gestation mouse embryos cultured for 16-40 hr exhibit morphological development comparable to embryos developing in utero.

Generating Neural Progenitors from Embryonic Stem Cells in Serum-Free Monolayer Cultures

The video demonstrates a protocol that uses serum-free media to generate neural progenitors from mouse embryonic stem cells. The desired concentration of embryonic stem cells, suspended in serum-free media, is plated on gelatin-coated coverslips. At an optimal cell density, cells produce autocrine signaling factors, which, along with the neural differentiation-inducing factors in the serum-free media, trigger the differentiation of embryonic stem cells into neural progenitors.
