Stereotaxic Transplantation

Stereotaxic transplantation is a neurosurgical technique that places cells, tissue, or therapeutic materials into precisely defined locations within the brain or spinal cord. Using a stereotaxic frame or image-guided system, researchers establish three-dimensional coordinates and advance a delivery instrument through a controlled trajectory to deposit the transplant at a targeted site while limiting damage to surrounding structures. This method supports studies of neural development, circuit repair, disease mechanisms, and cell-based therapies by enabling consistent implantation and subsequent assessment of graft survival, integration, and functional effects. It also provides a foundation for testing regenerative strategies in animal models of neurological disorders.

Source: Furmanski, O., et al. Controlled Cortical Impact Model of Mouse Brain Injury with Therapeutic Transplantation of Human Induced Pluripotent Stem Cell-Derived Neural Cells. J. Vis. Exp. (2019)In this video, human-induced pluripotent stem cell-derived neural cells are stereotaxically injected into the gray matter-white matter border of the deep cortex in a mouse model of traumatic brain injury. The transplanted cells integrate into the tissue, promoting functional recovery and circuit...

The monitoring of extracellular neurotransmitter levels in distinct brain regions of freely moving animals offers insights on the link between neurotransmitter release and behavior. In vivo microdialysis coupled with electrochemical detection provides excellent anatomical and chemical resolution; and information on how basal neurotransmission is altered by pharmacological or physiological manipulations.

Rodent Stereotaxic Surgery

Stereotaxic (or stereotactic) surgery is a method used to manipulate the brain of living animals. This technique allows researchers to accurately target deep structures within the brain through the use of a stereotaxic atlas, which provides the 3D coordinates of each area with respect to anatomical landmarks on the skull. After the skull is exposed, anesthetized animals are mounted on a specialized instrument known as a stereotaxic frame, which enables the precise placement of experimental...
