Ventral Basal Hypothalamus

The ventral basal hypothalamus is a region at the base of the brain that integrates internal physiological signals to coordinate endocrine, autonomic, and behavioral responses. Its neurons detect circulating hormones, nutrients, and neural inputs, then regulate pituitary activity through hypothalamic releasing factors, including signals delivered to the anterior pituitary through the hypophyseal portal system. This region contributes to energy balance, reproductive function, stress responses, and growth by linking body state with hormonal output and motivated behavior. Understanding its circuits and signaling pathways supports research into obesity, diabetes, infertility, stress-related disorders, and other conditions involving disrupted neuroendocrine regulation.

Despite the functional and medical importance of the hypothalamus, in utero genetic manipulation of its development has rarely been attempted. We show a detailed procedure for in utero electroporation into the mouse hypothalamus and show representative results of total and partial (regional) hypothalamic transfection.
