Reversible Irreversible Agents

Reversible and irreversible agents are pharmacological compounds classified by whether their binding to a molecular target can be undone, a distinction that helps predict drug duration, dosing, and recovery after exposure. Reversible agents typically interact through noncovalent forces and dissociate as concentrations change, whereas irreversible agents form exceptionally stable, often covalent bonds that permanently modify the target until new target molecules are produced. This classification is important in pharmacology for understanding enzyme inhibition, receptor regulation, toxicity, and therapeutic selectivity. Comparing these mechanisms supports drug design and helps researchers anticipate onset, persistence, reversibility of effects, and strategies for managing adverse responses.

The murine model of irreversible unilateral ureteric obstruction (UUO) is presented together with the model of reversible UUO in which the ureteric obstruction is relieved by anastomosis of the severed ureter into the bladder. This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.
