Aniline Derivatization

Aniline derivatization is a chemical labeling strategy that modifies carbonyl-containing biomolecules to improve their detection, separation, or structural analysis. In biochemical applications, aniline reacts as a nucleophile with an aldehyde, such as the reducing end of a carbohydrate, to form an imine; reductive amination can then stabilize the attached label as a secondary amine. The resulting derivatives often show enhanced ultraviolet or fluorescence response and improved ionization during mass spectrometry. This method supports glycan profiling, carbohydrate composition analysis, and quantitative studies of metabolites and other carbonyl-containing compounds.

Protocol for the Synthesis of Ortho-trifluoromethoxylated Aniline Derivatives

An operationally simple procedure for the synthesis of ortho-trifluoromethoxylated aniline derivatives via a two-step sequence of O-trifluoromethylation of N-aryl-N-hydroxyacetamide followed by thermally induced intramolecular OCF3-migration is reported.

Derivatization of Protein Crystals with I3C using Random Microseed Matrix Screening

This article presents a method to generate protein crystals derivatized with I3C (5-amino-2,4,6-triiodoisophthalic acid) using microseeding to generate new crystallization conditions in sparse matrix screens. The trays can be set up using liquid dispensing robots or by hand.

We describe a method protocol for the GC-based analysis of the aldonitrile acetate derivatives of glucosamine and muramic acid extracted from soil. For elucidation of the chemical mechanism, we also present a strategy to confirm the structure of the derivative and the ion fragments formed upon electron ionization.
