Diacylglycerol Lipase

Diacylglycerol lipase is an enzyme that hydrolyzes diacylglycerol, a membrane-associated lipid, to produce monoacylglycerol and a free fatty acid. It acts through cleavage of an ester bond in the diacylglycerol substrate, and specialized isoforms such as DAGLα and DAGLβ contribute to the formation of 2-arachidonoylglycerol, a major endocannabinoid. In biochemistry, studying diacylglycerol lipase helps clarify how lipid metabolism regulates intracellular signaling, synaptic communication, and inflammatory responses. Its activity and inhibition are also important for investigating endocannabinoid pathways and their potential relevance to neurological and metabolic disease research.

Many predicted (phospho)lipases are poorly characterized with regard to their substrate specificities and physiological functions. Here we provide a protocol to optimize enzyme activities, search for natural substrates, and propose physiological functions for these enzymes.

We describe a Flippase-induced intersectional Gal80/Gal4 repression (FINGR) method, allowing tissue-specific FLP to determine Gal80 expression patterns. Wherever Gal4 and FLP overlap, Gal4 expression is turned on (Gal80 flipped out) or off (Gal80 flipped in). The FINGR method is versatile for clonal analysis and neural circuit mapping.

This work presents a method of high-throughput screening using a universal genetic enzyme screening system that can be theoretically applied to over 200 enzymes. Here, the single screening system identifies three different enzymes (lipase, cellulase, and alkaline phosphatase) by simply changing the substrate used (p-nitrophenyl acetate, p-nitrophenyl-β-D-cellobioside, and phenyl phosphate).
