Huntingtin Protein

Huntingtin protein is a large, widely expressed protein involved in cellular organization, transport, and neuronal function, making it important in biochemistry and disease research. Its precise molecular mechanism remains complex, but huntingtin interacts with proteins that regulate vesicle trafficking, cytoskeletal dynamics, autophagy, and gene expression; an expanded polyglutamine region caused by CAG-repeat mutations can alter these interactions and promote toxic protein aggregation. Studying huntingtin structure, processing, and binding partners helps explain how altered protein homeostasis damages neurons in Huntington’s disease. This research supports the development of biomarkers and therapeutic strategies aimed at reducing mutant huntingtin or restoring its normal cellular functions.

The present protocol describes a procedure to perform fluorescent size exclusion chromatography (FSEC) on membrane proteins to assess their quality for downstream functional and structural analysis. Representative FSEC results collected for several G-protein coupled receptors (GPCRs) under detergent-solubilized and detergent-free conditions are presented.
