Primary 3d Cultures

Primary 3D cultures are laboratory-grown cell systems established directly from freshly isolated tissue and maintained in a three-dimensional environment, providing a model that can better preserve native cellular properties than conventional two-dimensional cultures. Cells grow within extracellular matrix materials or supporting scaffolds, where cell-cell contacts, cell-matrix interactions, nutrient gradients, and biochemical signaling shape organization and function. In biochemistry, these cultures help researchers examine tissue-specific metabolism, protein activity, signaling pathways, and responses to drugs or environmental conditions. Their closer resemblance to in vivo tissue makes them valuable for disease modeling, compound testing, and studying how cellular behavior changes during culture.

Vestibular Schwannomas (VSs) are non-malignant tumors of Schwann cell (SC) origin, associated with mutations in the NF2 tumor suppressor gene. We report a reproducible, efficient protocol for primary human VS cell culture that allows for molecular and cellular experimental manipulation and analysis and recapitulates the heterogeneous nature of human disease.
