Protein Expression Levels

Protein expression levels describe the amount of a specific protein produced and maintained in a cell, tissue, or biological sample, providing a key indicator of cellular function and regulation. They arise from coordinated control of gene transcription, messenger RNA processing and stability, translation by ribosomes, and protein folding, modification, localization, and degradation. In biochemistry, researchers measure expression levels using approaches such as immunoblotting, enzyme-linked immunosorbent assays, mass spectrometry, or fluorescent reporters. Comparing these levels across conditions can reveal pathway activity, responses to stimuli, disease-associated changes, and the effects of genetic or pharmacological interventions.

Quantitative Western Blotting to Estimate Protein Expression Levels

2025

The video demonstrates a total protein normalization-based western blotting technique for quantifying the target protein expression in a sample. By utilizing the total protein staining of the sample, a ratiometric analysis is performed to determine the relative expression level of the target protein.
