Automated Wounding Device

Automated Wounding Device is an instrument that creates reproducible injuries in cultured cell layers, enabling researchers to measure tissue repair and cell behavior under controlled conditions. In bioengineering, it applies a programmed mechanical stimulus to a confluent monolayer, producing wounds with consistent size and timing so closure can be tracked through imaging and quantitative analysis. This standardization supports studies of cell migration, proliferation, biomaterial performance, and engineered tissue repair while reducing operator variability and improving comparability across experiments and laboratories.

This protocol demonstrates how to achieve femto molar detection sensitivity of proteins in 10 µL of whole blood within 30 min. This can be achieved by using electrospun nanofibrous mats integrated in a lab-on-a-disc, which offers high surface area as well as effective mixing and washing for enhanced signal-to-noise ratio.

Here, an automated workflow to perform modular DNA "device" assembly using a modular cloning DNA assembly method on liquid-handling robots is presented. The protocol uses an intuitive software tool for generating liquid handler picklists for combinatorial DNA device library generation, which we demonstrate using two liquid handling platforms.
