Neutral Red Uptake Assay

The Neutral Red Uptake Assay is a colorimetric cell-viability test that estimates the number of living cells by measuring their ability to accumulate a dye. Viable cells actively transport neutral red across the plasma membrane and sequester it in acidic lysosomes, whereas damaged or nonviable cells take up less dye; after extraction, absorbance reflects cellular viability. In bioengineering, researchers use the assay to evaluate cytotoxicity, biomaterial and scaffold compatibility, and responses to drugs or engineered culture conditions. Its simple quantitative readout supports screening and optimization, although results depend on cell type, exposure conditions, and dye-handling steps.

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.

Here we present a cell-based flow cytometry method to detect neutralizing antibodies or other factors that interfere with the cellular uptake of enzyme replacement therapies in a human matrix, such as cerebral spinal fluid (CSF) or human serum.

Oil Red O Staining: A Technique for Staining Neutral Lipids in Hepatocytes for Detecting Hepatic Steatosis In Vitro

2025

In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.
